The link between paraquat exposure and Parkinson's disease is one of the most thoroughly studied associations in environmental neurology. This piece summarizes what the science actually shows — both the mechanism of injury and the human epidemiology.
Mechanism: oxidative stress and dopaminergic neurons
Paraquat is a redox-cycling compound. Inside cells it accepts an electron, becomes a free radical, and donates that electron to oxygen — producing superoxide and other reactive oxygen species. Dopaminergic neurons in the substantia nigra are unusually vulnerable to oxidative stress because of their high baseline mitochondrial demand and their reliance on dopamine, itself a redox-active molecule. Repeated paraquat exposure causes selective loss of these neurons — the same neurons whose progressive loss defines Parkinson's disease.
Mitochondrial dysfunction
Paraquat impairs complex I of the mitochondrial electron transport chain. Reduced ATP production and increased reactive oxygen species cause progressive neuronal injury. The MPTP model — a well-known experimental Parkinson's model — works through a similar mechanism, which is part of why researchers consider the paraquat-Parkinson's link mechanistically plausible.
Animal model evidence
Rodent studies have repeatedly shown that paraquat exposure produces selective loss of substantia nigra dopaminergic neurons with behavioral features consistent with parkinsonism. Combined exposure with maneb produces even larger effects, which is relevant for farmworkers exposed to multiple pesticides.
Human epidemiology
Case-control studies — including the Agricultural Health Study and multiple meta-analyses — have shown elevated odds ratios for Parkinson's disease among people with documented occupational paraquat exposure. Effect sizes are larger for higher cumulative exposure, consistent with a dose-response relationship.
Regulatory and scientific positions
The EPA continues to permit paraquat under Restricted Use rules. The International Agency for Research on Cancer has not formally classified paraquat as a Parkinson's risk factor — IARC focuses on cancer — but the National Institutes of Health and multiple academic centers describe the paraquat-Parkinson's association as well established.
